1、 Before operation: Prepare for shutdown and eliminate potential risks


1. Environmental and equipment inspection


Before starting the equipment, it is necessary to confirm that the environment where the entire biological cabinet is located is well ventilated and there is no airflow interference around (such as being away from air conditioning vents, doors and windows); Check whether the appearance of the cabinet is intact, the glass window rises and falls smoothly, and the countertop is free of damage or stains; Confirm the negative pressure status and wind speed parameters through the device display screen to ensure they meet the standards (generally, the average descending wind speed of the entire cabinet for Class II organisms is ≥ 0.38m/s, and the inflow wind speed is ≥ 0.58m/s). At the same time, turn on the ultraviolet toxic lamp and conduct a 30 minute or more toxic test on the cabinet environment (personnel are prohibited from staying during the toxic period).


2. Preparation of experimental materials


Clean the reagents and consumables required for the experiment (such as pipettes, culture dishes, centrifuge tubes) in advance, and place them in the order of "clean to contaminated": clean items (such as unopened consumables) are placed on the left side, the experimental operation area is located in the middle, and contaminated items (such as waste liquid tanks and toxic cotton pads) are placed on the right side to avoid cross movement of items; It is prohibited to bring personal items unrelated to the experiment (such as mobile phones, laptops) into the cabinet to prevent interference with airflow or pollution.


2、 In operation: strictly adhere to the "standardization" and control every detail at every step


1. Personnel protection and airflow stability


Operators need to wear personal protective equipment (PPE), including sterile gloves, protective clothing, goggles or face shields, and slowly enter the operating area after poisoning with their hands; Open the glass window to the specified height (usually 20-25cm, depending on the device identification), wait for the airflow to stabilize for 10-15 minutes before starting the experiment, to avoid sudden movement of objects that may damage the airflow barrier.


2. Core principles of experimental operation


During the operation, the arm should slowly extend from the front opening of the cabinet to avoid moving back and forth at the edge of the window; Maintain a vertical angle when using a pipette to prevent liquid droplets from contaminating the tabletop; If a sample leak occurs, immediately cover it with a tissue soaked in 75% ethanol or chlorine containing toxic agents, let it stand for 10 minutes, and clean it up to avoid the generation of aerosols during the cleaning process; Prohibit vigorous operations inside the cabinet (such as rapid shaking of centrifuge tubes, repeated opening and closing of container lids) to reduce the risk of biological hazard diffusion.


3. Management of Polluted Items


The waste liquid and consumables generated during the experiment should be promptly placed in a Chinese biological waste bag or high-pressure sterilization container, sealed and marked with a "biological hazard" label, and temporarily stored in the contaminated area on the right side of the cabinet. After the experiment is completed, they should be disposed of uniformly, and contaminated items are prohibited from being taken out of the cabinet.


3、 After operation: Establish a strong "closing checkpoint" to ensure a complete environment


1. Cleaning inside the cabinet and dealing with toxic substances


After the experiment, close the glass window and keep the fan running for more than 30 minutes to ensure that any residual aerosols in the cabinet are expelled; Subsequently, wipe the countertop, side walls, and glass windows inside the cabinet with 75% ethanol, focusing on cleaning the areas that have been in contact during the operation; If the experiment involves highly pathogenic microorganisms, additional formaldehyde fumigation or hydrogen peroxide atomization should be carried out to reduce toxicity. After poisoning, ventilation should be carried out until there are no residues.


2. Device shutdown and recording


After confirming the completion of cleaning the Xiao toxin, turn off the fan and lighting in sequence, record the experimental name, usage time, equipment status, and abnormal conditions (such as wind speed fluctuations and leakage events) of this operation for subsequent equipment maintenance and traceability.
